The first Stone audiences see Thanos obtain is the Space Stone, which has significance, as it was the first stone introduced in the Marvel Cinematic Universe, originally appearing as the Tesserect in Captain America - Il primo Vendicatore (2011). This movie confirms that the stone is actually housed inside the cube, something briefly suggested during Thor's vision in Avengers: Age of Ultron (2015).
