Contrary to popular belief, the bookcase falling down behind Peter, Ray and Egon was not an unscripted moment and was shot roughly according to the original script which can be found in the 1985 book "Making Ghostbusters" by Don Shay. Also according to the book, director Ivan Reitman decided on his way to the set that the library ghost sequence demanded a stronger setup and thus added Venkman collecting a slime sample and the stacked books to the mix.
