Due to Darth Vader's backstory being expanded in the rest of the original trilogy, the prequels and Star Wars: The Clone Wars (2008), Tarkin's quote (directed towards Vader), "You, my friend, are all that's left of their religion" now implies that he could have been aware about Vader's identity as Anakin Skywalker. Tarkin and Skywalker fought side by side in the Clone Wars.
