To prevent Alice and company from entering the hive, Wesker orders the release of cerberus. When they survive the creatures onslaught and make it to the front of the Hive's gate, only then he give the order to close it down. We see the huge and slow blast doors closing, slow enough that the protagonists just walk between them. If he had ordered to shut down the gate just at the beginning, Alice and friends would not be able to enter the Hive in the first place.
